The budget airline will operate twice week services to Rome and Newcastle from October.
Bookings for Newcastle are already on sale, while flights to Rome–Fiumicino International Airport will be on sale from tomorrow.
It follows the start of direct flights from Cork to Valencia in Spain last week.
Ryanair currently operates 25 of the 41 routes available from the airport.
Cork Airport say demand has been strong for a Cork-Rome connection for years and say the Cork to Newcastle route will suit fans of Premier League football.
